餐饮废水高效降解蛋白菌株的分离筛选及诱变选育研究

摘    要:从龙潭山宾馆厨房排污口分离得到11株产蛋白酶菌株,培养相同时间测定蛋白质降解率,从中筛选出一株蛋白质降解率高的菌株P4.通过基本的生理生化性质的测定,初步鉴定为巨大芽孢杆菌(Bacillus megaterium).通过紫外诱变提高菌株的产蛋白酶能力,在发酵时间为120 h的前提下,蛋白质降解能力约提高10.96%.利用正交试验确定蛋白质最佳降解条件:酵母膏最优浓度为2.5 g/L,葡萄糖最优浓度为5.0 g/L,Cu2+最优浓度为1.0 g/L,最优装样量为40%.

Abstract:The strain used in the experiment of Longtan Mountain Hotel kitchen outfall, screening, rescreening, getting better performance degradation strains. We measured the basic physiological and biochemical properties,growth curve, K2HPO4 and phenol highest tolerable concentration. Single factor experiments to determine the ootimum nitrogen source was venal extract the fermentation time is 120h under the premise 6%. Orthogonal test to determine optimal protein of 2.5 g/L, the optimal glucose concentration was 1.0 g/L, the optimal amount of sample loading 40%.
