摘    要:目的采用基质辅助激光解吸附电离飞行时间质谱法对金桔表面细菌进行筛查,初步掌握金桔表面细菌分布情况,为金桔的食用安全提供科学依据。方法以4种常见食源性致病菌(沙门氏菌、阪崎肠杆菌、单增李斯特菌、金黄色葡萄球菌)为目标菌,同时分离未知菌,通过基质辅助激光解吸附电离飞行时间质谱法进行鉴定。结果共从17个样品中分离到12种细菌,分别为产气肠杆菌、阴沟肠杆菌、蜡样芽孢杆菌、肺炎克雷伯菌、腐生葡萄球菌、木糖葡萄球菌、大肠埃希氏菌、生癌肠杆菌、解鸟氨酸拉乌尔菌、阪崎肠杆菌、分散团菌和恶臭假单胞菌。结论本方法能快速鉴定未知细菌,从金桔上分离到的细菌多数为条件致病菌,存在一定的食用风险。

Abstract:Objective Preliminary study on the distribution of bacteria on the surface of kumquat and provide scientific basis for the safety of kumquat.Method Several common food-borne pathogens (Salmonella, Enterobacter sakazakii, Listeria monocytogenes, Escherichia coli O157, Staphylococcus aureus) were used as target bacteria, and unknown bacteria were isolated which identified by matrix assisted laser desorption/ionization time-of -flight mass spectrometry.Result Twelve species of bacteria were isolated from 17 samples, including Enterobacter cloacae, Bacillus cereus, Klebsiella pneumoniae, Staphylococcus saprophyticus, Staphylococcus aureus, Escherichia coli, Enterobacter spp. Ornithine Laurus, Enterobacter sakazakii, Dispersive and Pseudomonas putida.Conclusion Unknown bacteria could identified quickly by Matrix-assisted laser desorption ionization time-of-flight mass spectrometry , most of the bacteria isolated from kumquat in this experiment were opportunistic pathogens, which leaded to a certain food risk.
