小叶丁香的悬浮细胞培养研究

摘    要:小叶丁香(Syringa pubescens Turcz.)为木犀科(Oleaceae)丁香属(Synnga)植物,其中主要药用有效成分为属于裂环环烯醚萜苷类化合物的橄榄苦甙.选择小叶丁香的叶、茎、芽为外植体,在MS和LS等培养基上进行愈伤组织诱导与培养,并在继代3次以后转入悬浮细胞培养,测定了悬浮培养的细胞生长周期,首次建立了小叶丁香的悬浮细胞培养体系;采用RP-HPLC方法测定悬浮细胞培养物和培养液中橄榄苦苷的含量,并且利用1HNMR和13CNMR确定其结构.

Abstract:Syringa pubescens Turcz. is a kind of shrub widely found in China.Oleuropein, a secoiridoid glucoside, is the primary chemical constituent in this plant with an active potential medicine. Some types of calli were induced and cultured from the cotyledon, the spire, and the caulicle of Syringa pubescens Turcz. on LS or MS culture mediums in this paper.After three times of subculture periods, the calli are transferred into the suspension cell culture.Growth curve of the suspension cell culture is determined.The suspension cell culture system of Syringa pubescens Turcz.is established on this plant for the first time.The content of oleuropein from the callus cultured cell is determined by RP-HPLC and its chemical structure is elucidated by~()~1HNMR and~()~(13)CNMR. The experiment provides data for the production of oleuropein by the tissue (cell) culture.
