大兴安岭金莲花粗多糖提取工艺研究

摘    要:以大兴安岭金莲花为原料,采用水提醇沉法研究了大兴安岭金莲花粗多糖的提取工艺及苯酚-硫酸法测定金莲花粗多糖含量.在单因素实验的基础上,通过正交实验确定了浸提时间、浸提温度、料液比、乙醇浓度对大兴安岭金莲花中粗多糖浸提效果的影响及最佳提取条件.结果表明:浸提温度80℃,浸提时间为3 h,料液比1 60,乙醇浓度为85%的条件下,大兴安岭金莲花粗多糖得率最高,可达29.63 mg/g.

Abstract:Trollius Chinensis Bunge in Daxing’an Mountain was used as raw material,the extraction factors of crude polysaccharide were investigated by water extraction and alcohol precipitation methods and the crude polysaccharide was determined by the phenol-sulfuric acid reaction.On basis of single-factor test,the effects such as ratio of material to water,immersion time,immersion temperature and ethanol concentration were determined and the optimum conditions was obtained by orthogonal test.The results showed that the optimized condition of immersion temperature is 80 ℃,immersion time is 3 h,the ratio of material to water is 160,the concentration of alcohol is 80%.Under the optimal conditions,the crude polysaccharide from Trollius Chinensis Bunge in Greater Khingan Range was up to 29.63 mg/g.
