大豆愈伤组织的诱导与离体培养

摘    要:本研究选用6个大豆品种,以大豆下胚轴和子叶为供试外植体,探讨了激素、基因型及外植体的差异对形成愈伤组织的影响,结果表明,诱导愈伤组织合适的培养条件是:MS+2,4-D2.0mg/L+6-BA0.5mg/L+蔗糖3%+琼脂0.8%,所有的基因型均能形成愈伤组织,但不同基因型诱导愈伤组织能力有差异,以中黄十三最高;不同外植体诱导愈伤组织能力也不一样,下胚轴的诱导能力比子叶强。

Abstract:The six soybeans genotypes were studied in this experiment.Hypocotyls and cotyledons were used as explants to study the effect on the difference of phytohormone,genotype and explants concentration for soybean callus formation.The most desirable medium in the test was MS+2,4-D2.0mg/L+(6-BA0.5mg/L)+sucrose3%+agar0.8%.All the genotypes used in the test produced calli,but rate of callus formation varied between the genotypes,the rate of zhonghuang13 was the highest.The rate of callus formation varied from different explants.The hypocotyls had higher callus induction than cotyledons.
