牛蒡子苷及苷元的提取和纯化

摘    要:采用醇提和硅胶柱层析的方法从牛蒡子中分离和纯化得到有效成分牛蒡子苷和牛蒡子苷元。通过L,(4^3)正交实验发现牛蒡子提取的最佳条件为:乙醇浓度80%,回流3次,回流时间1h,加8倍溶剂量。使用硅胶柱层析分离方法纯化牛蒡子苷和苷元,依次用二氯甲烷:甲醇20:1,二氯甲烷:乙酸乙酯:甲醇4:4:1,二氯甲烷:乙酸乙酯5:4梯度洗脱,最后得到牛蒡子苷和牛蒡子苷元的白色粉末,经检测.两者的纯度分别为95.5%和94%。

Abstract:Arctiin and Arctigenin's active components in Fructus Arctii, was extracted with ethanol and separated from the extraction liquid with silicagel column adsorption chromatography. The extraction procedure was optimized using Lν9 (4^3) orthogonal array design. The optimum technological parameters for separating Fructus Arctii were determined as follows: ethanol concentration 80%, reflux time 1 h, 8 times solvent quantity. Arctiin and Arctigenin were purified by silicagel column chromatography with gradient elution successive using dichloromethane :methnol (20:1, v/v), dichloromethane:ethyl acetate:methnol (4:4:1, v/v/v) and dichloromethane: ethyl acetate (5:4, v/v). White power of arctiin and arctigenin were gained and determined by HPLC. The content of arctiin and arctigenin were 95.5% and 94% respectively.
