摘    要:从多个方面测定、比较Bio-sep CL6B与Sepharose CL-6B两种商品化交联琼脂糖凝胶的性能。采用不同分辨率显微镜观察凝胶外观形态、进行耐热试验、分析有机元素及金属元素含量,以及测定二者耐压、流速、色谱特性等。结果表明,两种产品外观均为乳白色球状颗粒,球形形态完好,粒径范围在30~150μm具有较好的分布;均可耐受120℃下30 min热处理;有机元素碳、氢、氮以及其它微量金属元素含量相近;两种产品最大耐压都在80 kPa以上,最大流速可达7.0 mL/min。分离特性测定表明,两种产品色谱分离特性基本相同,对分子量在104~106道尔顿之间的蛋白能实现较好的分离。

Abstract:The characteristics of two kinds of cross-linked agarose gel in industrial applications like Bio-sep CL 6B and Sepharose CL 6B were compared in different aspects.The physicochemisty characteristics of the two cross-linked agarose gels including particle shape,particle size distribution,heat stability,elementals and metals levels,back-pressure,flow-rate,chromatographic properties,etc,were studied.The results showed that the appearance of the both products were white globose particles.With the particle size between 30 and 150 μm,and both of them can withstand heating for 30 min at 120 ℃.The content of carbon(C),hydrogen(H),oxygen(O) and other microelement of the two products were nearly the same.The biggest pressure that the two products can suffer was above 80 kPa,the fastest flow-rate was 7.0 mL/min.The chromatographic separate properties of the two products were basically the same,by which the protein that molecular weight between 104 and 106 can be well separated.
