银杏萜内酯超临界CO2萃取结晶前处理工艺研究

摘    要:本文根据中草药有效成分含量低、超临界稀溶液特点及溶剂提取技术原理,以银杏叶粗提物为原料,通过单因素实验的方法,探讨液固比、浸提温度、浸提时间、提取次数对超临界CO2萃取结晶银杏萜内酯前处理工艺的影响。实验结果表明:在45℃水浴条件下,以20倍银杏叶粗提物体积的混合溶剂(40%丙酮-60%石油醚),浸提2次,每次4h,银杏萜内酯前处理效果较佳,萜内酯浸出率在95%以上,浸出物内酯含量在16%以上。采用高效液相色谱法测定银杏内酯含量准确,重现性好。

Abstract:Based on low content of active ingredients of Chinese herb, and the principle of supercritical dilute solution extractionand the fundamental principle of solvent extraction, we prepared the extract of ginkgo biloba L. as experimental material toinvestigate the effect of ratio of liquid to solid material, extraction temperature and extraction time on pre-treatment of supercriticalCO2 extracting and crystallizing ginkgolides via single factor experiments. The results showed with that 20 times more volumesolvent (40% acetone-60% ether) the optimum conditions were: 45℃ extraction temperature, extraction 2 times and 4h everytime and ratio of extraction more than 95%, the ginkgolide yield was more than 16%. The method of determination was extractand steady.
