摘    要:采用现代生物酶解技术制备羊胎盘生物活性肽 ,以水解度 (DH)和酸溶性肽得率(YASP)为指标 ,在单酶水解的基础上探索更加有效的新工艺———双酶水解工艺。实验确定了木瓜蛋白酶 (Papain)与胰蛋白酶 (Trypsin)的等活力混合酶为水解羊胎盘的最佳双酶组合 ,其最佳酶解工艺条件为 :pH7 0、温度 5 2℃、酶活添加量 3 0 0 0u/ g(料 )、底物浓度 10 %、水解时间 3h ,酶解前最佳热处理条件为沸水浴 15min。

Abstract:Bioactive peptide was prepared by enzymatic hydrolysis of proteins from sheep placenta. By measuring the degree of hydrolysis and the yield of acid-soluble peptide, bienzyme hydrolysis was studied on the basis of the experimental results of mono-enzyme hydrolysis. The mixture of Papain and Trypsin with the same activities was selected as the optimum hydrolytic condition,and other optimum parameters of the bienzyme hydrolysis were:the pH was 7.0,the reaction temperature was 52 ℃,the enzyme activity was 3?000U/g (sheep placenta) and the substrate concentration was 10%,the reaction time was 3 hours.The optimal heating condition was 100℃ for 15 minutes.
