木醋杆菌的培养基优化研究

摘    要:采用六因素三水平正交实验对木醋杆菌Acetobacter xylinum HN001的培养基进行了优化,确定了最佳培养基组成为:葡萄糖30 g/L,蛋白胨7.5 g/L,酵母粉10 g/L,磷酸氢二钠7.5 g/L,柠檬酸0.5 g/L,pH 5.0,30℃。采用优化的培养基制备细菌纤维素,应用傅立叶红外光谱、热重分析和扫描电子显微镜对其结构和性能进行表征。红外光谱测试显示含有纤维素的特征吸收峰。热重分析结果表明细菌纤维素最大失重温度为319.06℃,热稳定性较好。扫描电镜观察到细菌纤维素的纳米网状结构。

Abstract:The medium optimization of Acetobacter xylinun HN001 was obtained by orthogonal experimental design.The culture media of Acetobacter xylinun HN001 was optimized as:Glucose 30 g/L,peptone 7.5 g/L,yeast extract 10 g/L,Na2HPO4 7.5 g/L,Citric acid 0.5 g/L,pH 5.0,30℃.In this study,bacterial cellulose was synthesized by incubation of Acetobacter xylinun HN001 in the modified culture medium.And they were characterized by FT-IR spectrum,scanning electron microscope(SEM) and thermo gravimetric analyzer.The characteristic adsorption peaks of bacterial cellulose(BC) were in FTIR spectra.The maximum weight loss temperature of BC was 319.06℃,thermal stability of BC was better.Nanostructure of BC was observed by SEM.
