甲硝羟乙唑衍生物(CM)对离体乏氧L_(7712)翻胞的辐射增敏作用

摘    要:本文报道了用马血清代替小牛血清改进和建立了能用于检测L_(7712)细胞存活率的ADC法。并用此法研究证明了我们合成的甲硝羟乙唑衍生物(CM)对离体乏氧L_(7712)细胞有明显的辐射增敏作用,其SER值为2左右,较原化合物灭滴灵的SER(1.2)约提高67%。CM对缺氧细胞存活率的抑制作用比有氧下强。与前文报道的CM对荷瘤小鼠S_(180)缺氧下照射有辐射增敏作用及对鼠乏氧S_(180)生长有抑制作用的结果一致。作者对台盼蓝染色法和ADC法检测细胞存活率作了比较。

Abstract:The modified ADC method which can be used in measuring the sur-vival fraction of L_(7712) cells was established using horse serum instead of calf serum.It demonstrated the obvious radiosensitizing effect of metronidazole derivative (CM)on in vitro bypoxic L_(7712) cells. Its SER was about 2, and was about 67 percent hig-her than SER (1 .2) of the original compound, metronidazole. CM was found to bestronger under hypoxic condition than under aerated condition in the inbibition of cellsurvival fraction. It was consistent with the results of the preceeding paper that CMshowed a strong ability to sensitize sarcoma 180 to radiation and to inhibit the growthof sarcoma 180 under hypoxic condition. Using trypan blue staining method and theADC method to observe cell survival were compared.
