| 摘 要： | 为比较不同类型烟叶中游离二肽的组成以及含量的差异,利用甲醇/三氟乙酸溶液提取,Fmoc 衍生化-液质联用分析烟叶中的游离二肽。结果显示:1)通过对二肽在质谱中断裂模式的探讨,共鉴定出烤烟烟叶中14 种、白肋烟中11 种、香料烟中9 种游离二肽。游离二肽的含量范围为1μg/g-120μg/g。2)不同类型烟叶在二肽种类上有一定程度的差异,谷氨酰胺(赖氨酸)-苯丙氨酸(KF/QF)以及双丙氨肽(AA)是三种类型烟叶中共有的含量较大组分,烤烟中脯氨酰缬氨酸(PV)以及甘氨酰谷氨酰胺(赖氨酸)(GQ/GK)含量较高,白肋烟中天冬氨酰组氨酸(DH)含量较高,而香料烟中缬氨酰苯丙氨酸(VF/FV)以及谷氨酰谷氨酰胺(赖氨酸)(EK/KE)这两种二肽含量较高。

| Abstract: | In order to compare the differences in composition and content of free dipeptides among various tobacco types, free dipeptides in tobacco leaves were separated and analyzed by liquid chromatography coupled with mass spectrometry after methanol/TFA extraction and Fmoc derivatation. Test results showed that (1) 14 dipeptides in flue-cured tobacco, 11 in burley tobacco and 9 in oriental tobacco were identified through fragmentation mechanism discussion. Dipeptides content in tobacco was among 1μg/g-120μg/g. (2) Dipeptides of KF/QF and AA had high concentration in all tobacco types. Flue-cured tobacco samples had high dipeptides of PV and GQ/GK while burley samples were high in DH and oriental tobacco was high in VF/FV and EK/KE. |
