酶法提取茶树菇多肽工艺及体外抗氧化研究

摘    要:试验用酶法提取茶树菇多肽,考察不同料液比、加酶量(质量分数)、提取温度以及提取时间对茶树菇多肽提取量的影响。通过清除DPPH自由基、总还原力测定研究茶树菇多肽体外抗氧化作用,并与Vc和BHT的抗氧化作用进行比较。试验结果显示,茶树菇多肽提取工艺为料液比1:60,2.0%复合蛋白酶,提取温度45 ℃,提取时间3.0 h。在此工艺条件下,多肽提取量最高,达到93.17 mg/g。体外抗氧化试验结果显示,当茶树菇多肽浓度为1.5 mg/mL时,对DPPH自由基清除率为44.8%,D700 nm达到0.55。说明茶树菇多肽对DPPH自由基有一定的清除率,有较好的总还原力。

Abstract:With Agrocybe aegirit as raw material,polypeptides were extracted by enzymatic method.The effect of differentratio of material to liquid,enzyme amount (concentration),extraction temperature,and extraction time on the polypeptides yield were investigated.The antioxidative effect of Agrocybe aegirit polypeptide in vitro was studied by scavenging DPPH free radical and total reducing power,and compared with that of Vc and BHT.The results showed that the best extraction technology was 1:60,2.0% compound protease,45 ℃,3.0 hours.Under this condition,the amount of polypeptide was 93.17 mg/mL.When the concentration of polypeptides is 1.5 mg/mL,the clearance for DPPH radical reach 44.8%,and D700 nm reach 0.55.The results of antioxidant test in vitro showed that the peptide of Agrocybe aegirit had a certain scavenging rate to DPPH free radical and a strong total reducing power.
