酱香型白酒中非乙醇物质抗幽门螺杆菌代谢物诱导的胃黏膜上皮细胞炎症作用研究

摘    要:目的:研究酱香型白酒中非乙醇物质(JFY)抗幽门螺杆菌代谢物(Helicobacter pylori Metabolites,HPM)导致的胃黏膜上皮细胞(GES-1)炎性损伤作用。方法:参考文献方法获得JFY及HPM;MTT法检测JFY、HPM对胃黏膜上皮细胞(GES-1)的细胞活性的影响;ELISA测定JFY对相关炎症因子白细胞介素-1β(Interleukin-1β,IL-1β)、白细胞介素-8(Interleukin-8,IL-8)及肿瘤坏死因子-α(Tumor Necrosis Factor-α,TNF-α)等的影响;Western blotting检测Bax、Bcl-2和NF-κB信号通路相关蛋白的表达,探索JFY抗HPM刺激GES-1诱发凋亡的作用机制。结果:HPM显著降低GES-1细胞活性;JFY预处理GES-1细胞显著抑制由HPM刺激导致的细胞活性降低(P>0.05),JFY抑制HPM刺激导致的IL-1β、IL-8及TNF-α分泌增加。Western blotting结果显示JFY可通过调节GES-1细胞内Bax、Bcl-2及NF-κB信号通路蛋白的表达发挥抗炎作用。结论:酱香型白酒中含有的生物活性物质具有一定的抗炎活性,可减弱HPM对GES-1细胞损伤,抑制炎症因子IL-1β、IL-8及TNF-α的分泌及调控Bax、Bcl-2及NF-κB信号通路蛋白的表达发挥抗炎作用。

Abstract:Objective:To study the anti-inflammatory potency of bio-active substance in Moutai-flavor liquor in human gastric epithelial cells induced by Helicobacter pylori metabolites.Methods:HPM and JFY were obtained via the method reported previously;The inflammatory factor expression was determined by ELISA;The protein expressions of NF-κB pathway,Bax and Bcl-2 were detected by Western blotting.Results:JFY significantly inhibited HPM-induced decrease in cell viability (P>0.05),JFY decreased the secretion of IL-1β,IL-8 and TNF-α in GES-1 cells induced by HPM stimuli.JFY inhibited HPM-induced GES-1 inflammation by reducing the expression of Bax,increasing the expression of Bcl-2,and blocking the NF-κB pathway activation.Conclusion:The JFY played a significant anti-inflammatory role by inhibiting the secretion of inflammatory factors,and regulating the expressions of Bax,Bcl-2,and NF-κB pathway.
