石榴酸对人膀胱癌T24细胞凋亡的影响

摘    要:利用MTT法检测石榴酸对T24细胞增殖的影响,利用Hoechst33342染色后观察细胞形态学的变化,并用流式细胞仪检测细胞凋亡率。MTT分析结果显示,石榴酸的半致死浓度(IC50)为30μmol/L。应用荧光显微镜观察Hoechst荧光染色后细胞核形态变化,石榴酸处理T24细胞后,细胞核体积明显变小,并呈现皱缩致密的颗粒状的强荧光。应用流式细胞仪分析石榴酸对细胞凋亡的影响,发现石榴酸能够诱导膀胱癌T24细胞的凋亡,30μmol/L石榴酸处理细胞24h,凋亡率为49.4%。说明石榴酸能够显著抑制膀胱癌T24细胞的活力,有效诱导T24细胞凋亡。

Abstract:Effect of punicic acid on the apoptosis of bladder carcinoma T24 was investigated by MTT analysis,including the cell viability of T24 cell lines with different concentrations and various treatment periods.Results showed that punicic acid inhibited proliferation of bladder carcinoma T24 in a dose-dependant manner with IC50 of 30 μmol/L and fluorescence microscopy after Hoechst33342 staining showed obvious morphological changes of T24 cell apoptosis.Flow cytometry analysis showed that punicic acid induced apoptosis in bladder carcinoma T24.The rate of apoptosis was 49.4% after treatment of 30 μmol/L punicic acid for 24 h.It was confirmed that punicic acid inhibit proliferation of bladder carcinoma T24 and induced apoptosis in bladder carcinoma T24.
