维药新塔花中总黄酮纯化工艺研究

摘    要:以新疆特有的芳香新塔花为研究对象,通过用大孔树脂对提取的总黄酮进行吸附与解吸,对大孔树脂进行筛选,确定比较理想的大孔树脂。采用吸附率最大和解析率最大的大孔树脂对芳香新塔花总黄酮其他纯化工艺参数进行考察,包括上样浓度、大孔树脂梯度洗脱、径高比、上样液流速的影响。最佳纯化工艺为:大孔树脂型号为HPD-600,上样液浓度为4.70mg/mL,径高比为1:10,七样液流速为0.5mL/min,洗脱时合并30%vol、50%vol、70%vol的乙醇洗脱液。

Abstract:In this paper the ideal macroporous resin is defined by using it to absorb and desorb distilled flavonoids and then sieve macroporous resin with the Ziziphora as the research object. Then other parameters of purification technology of total flavonoids in Ziziphora were investigated by using macroporous resin with high absorption rate and resolution factor, which in terms of influence of the concentration of sample, gradient elution of macroporous resin, diameter-height ratio, and the speed of the concentration of sample. The final technology was as follows: the type of macroporous resin HPD-600, the concentration of sample 4.70mg/ml, elution liquid of 30%vol, 50%vol and 70%vol. The diameter-height ratio was 1:10. The speed of the concentration of sample was 0.5ml/min.
