Abstract:Waters Corporation (NYSE; WAT) has introduced a novel automated high performance liquid chromatography (HPLC) system of drug researchers involved in disease target identification and pharmacokinetic studies of biomolecule and small molecule drug candidates. The system which features a sub-20μL internal volume and patented light-guided flow cell offers 50X or better mass sensitivity than traditional HPLC on subpicomole quantities of biomolecules.The CapLC System is a product of years of research and development. It is the first single-vendor-integrated LC system optimized for capillary-scale LC/MS separations. One of is most significant inno- vations is a patented light-guided flow cell that employs state-of-the-art fiber optic technology to enhance sensitivity. The flow cell is made of a unique fluoropolymer that enhances signal-to-noise so that scientists will be able to obtain sensitive and linear results over a wide dvnamic range. Results are comparable or superior to traditional HPLC in terms of retention time reproducibility and resolution. The CapLC System's accurate solvent delivery capability ensures reproducible isocratic and gradient performance, even at the low μL flow rates that are essential for obtaining quality electrospray mass spectra. The CapLC Sytem's low-flow-rate solvent delivery is so precise it can deliver the entire sample into the mass detector without splitting the flow. Analytical HPLC systems converted to capillary LC work must divert some of the sample to waste ahead of the mass detector so as not to overwhelm it. Most chemists are (more) fundamentally opposed to wasting any sample or solvent, but have had few options for acquiring mass spectra on such small samples.Equipped with an X,Y,Z sample manager and photodiode array detector,the system is a complete automated capiuary LC/MS system package. Automating assays and conserving bench space are major hot buttons with pharmaceutical companies striving to contain costs while increasing the number of drug leads they screen and investigate.The CapLC System is engineered specifically for use with Waters ZMD mass spectrometers and others made by Micromass Ltd (Manchester, England) including the Micromass橪CT Quattro橪C, Quattro Elite? and Q-TOF? Sotware control and data acquisition for the CapLC System is provided by Micromass MassLynx?Software which is Y2K-compliant.
