中国林蛙皮抗菌肽的提取纯化及抑菌活性检测

摘    要:采用酸化甲醇法提取中国林蛙皮肤中的抗菌肽,并采用凝胶色谱的方法对提取物进行纯化。结果表明:采用Sephadex G-50 和Sephadex G-25 串联凝胶层析法获取的F2 组分,即纯化后的抗菌肽的抑菌率可达90% 以上。抗菌肽对革兰氏阴性菌和阳性菌以及真菌均有一定抑制效果。对各受试菌株的最小抑菌浓度(MIC)分别为:金黄色葡萄球菌和链球菌为0.2mg/mL;枯草芽孢杆菌和大肠杆菌为0.4mg/mL;蜡样芽孢杆菌为0.8mg/mL;白色念珠菌为1.2mg/mL;酿酒酵母和毛霉为1.5mg/mL;根霉为2.0mg/mL。

Abstract:Rana chensinensis skin was extracted using acidified methanol for preparing anti-bacterial peptides and the resulting crude extract was then purified by gel filtration chromatography.Purified anti-bacterial peptides were found to be rich in F2 fraction up to 90% by tandem Sephadex G-50 and G-25 column chromatography.The purified anti-bacterial peptides could effectively inhibit the growth of Gram-positive bacteria,Gram-negative bacteria and fungus.The minimum inhibitory concentra-tions(MIC) were 0.2 mg/mL a...
