Effects of temperature and juvenile hormone analogs on the emergence time and rate of Aphidius gifuensis (Hymenoptera: Aphidiidae)

Abstract:  Background  The aim of this study is to determine the effects of juvenile hormone analogs and temperature on the emergence rate and emergence time of Aphidius gifuensis pupa. The optimal combination of juvenile hormone analogs and temperature that can maintain a high survival rate and delay the emergence time of A. gifuensis pupa was obtained, in order to provide basis for the large-scale production and release of A. gifuensis.  Methods  Aphidius gifuensis were treated with control (10% acetone) and juvenile hormone analogue by liquid immersion, and then placed in an artificial climate chamber with relative humidity of 75%±5% and photoperiod of 12L: 12D at different temperatures (10℃, 16℃, 22℃, 32℃, 38℃). The emergence time, total number of aphid mummie and number of adults were observed.  Results  (1) The emergence time of A. gifuensis could be effectively delayed under specific combination of juvenile hormone analogs and temperature. Under the combinations of 10℃+ 1000 ng/μL ZR-512, 22℃ + 1000 ng/μL ZR-512 or 200 ng/μL ZR-777, the emergence time of A. gifuensis pupa were delayed by 80.00h, 40.00 and 32.00 h, respectively. However, after treated with ZR-512(5000 ng/μL), the emergence time was gradually decreased with the increase of temperature. (2) juvenile hormone analogs and temperature had a significant influence on the emergence rate of A. gifuensis pupa: Under the combination of 10℃+ 1000 ng/μL ZR-512, the emergence rate of A. gifuensis pupa was improved about 80.00%. The high temperature had a negative effect on the emergence rate and emergence time of A. gifuensis pupa. When the temperature was 38℃, none of the pupae emerged.  Conclusion  Under the combination of temperature 10℃ + juvenile hormone analogs of 1000 ng/μL ZR-512, the emergence time can be effectively delayed and emergence rate of A. gifuensis pupa can be increased significantly. Such combined treatment has a good application value in large-scale reproduction and preservation of A. gifuensis in production.
