大孔吸附树脂选择性提取黄柏生物碱工艺研究

摘    要:探讨了利用大孔树脂吸附分离技术将黄柏中小檗碱、掌叶防己碱、药根碱进行选择性的提取方法.通过考察上样液浓度和上柱流速对三种生物碱动态吸附量的影响,确定了较优的吸附条件为:上样质量浓度20.00 mg/mL,上柱流速3 mL/min;利用均匀设计法优化了洗脱条件,确定了较优洗脱条件为:用体积百分比为78%乙醇溶液作洗脱液,洗脱液用量与上样量的体积比0.04:1,洗脱流速1.5 mL/min.以此条件对黄柏水提液进行处理,3种生物碱的收率较高且相对含量的与黄柏水提液最接近,该方法能够对黄柏中的小檗碱、掌叶防己碱、药根碱进行有选择性提取富集,具有一定的实际应用价值.

Abstract:The method is discussed using macroporous adsorbent resin adsorption separation technology to distil berberine, palmatine and jateorrhizine selectively. By observing the influence of loading liquid concentration and flow velocity to these three alkaloids dynamic adsorption amount, optimization of adsorption conditions confirmed are loading concentration 20. 00 mg/mL and flow velocity 3.0 mL/min ; using uniform design to optimize elution conditions, and optimization of elution conditions are as follows : the concentration of ethanol in the elution solution is 78% (V/ V), the volume ratio of eluant amount and loading amount is 0. 04 : 1, the flow velocity of elution is 1.5 mL/min. In the condition, cortex phallodendri water extract is disposaled, these three alkaloids' adsorption ratios are relatively high and their relative contents are approached with cortex phallodendri water extract. This method can distil the enrichment of berberine, palmatine and jateorrhizine selectively and have certain significance of application.
