摘    要:目的:为探索花状纳米金(Gold nanoflowers,AuNFs)在免疫层析方法中应用的可行性,并探究其在果蔬样品中的应用。方法:使用AuNFs、球状金(Gold nanospheres,AuNSs)分别和苯醚甲环唑单克隆抗体制备免疫探针。优化反应体系,构建苯醚甲环唑(Difenoconazole,DIFE)免疫层析试纸条(immunochromatographic strip,IGTS)。验证该试纸条的检出限、特异性、准确性和精密度,并对果蔬样本进行检测。结果:以AuNFs为探针制备的试纸条检出限为0.05 mg/kg,检测限比以AuNSs为探针制备的试纸条检测限提高10倍,检测时间为15 min,与其他三唑类农药无交叉反应,假阴性率为0,假阳性率为3.75 %,稳定性良好。结论:以AuNFs为探针制备的试纸条性能良好,可以有效提升检测限,可准确检测果蔬中苯醚甲环唑含量是否超标。

Abstract:To explore the feasibility of Gold nanoflowers (AuNFs) in immunochromatography and its application in fruit and vegetable samples. Methods: Immune probes were prepared using AuNFs, Gold nanospheres (AuNSs) and monoclonal antibodies against difenoconazole, respectively. The reaction system was optimized to construct the immunochromatographic strip of difenoconazole. The strip''s detection limit, specificity, accuracy and stability were verified, and the fruit and vegetable samples were tested. Results: The detection limit of AuNFs test strip was 0.05 mg/kg, the sensitivity of AuNSs test strip was 10 times higher than that of AuNSs test strip, the detection time was 15 min, there was no cross reaction with other triazole pesticides, the false negative rate was 0, the false positive rate was 3.75%, and the stability was good. Conclusion: The test strip prepared with AuNFs as probe has good performance, can effectively improve the sensitivity, and can accurately detect whether the content of Difenoconazole in fruits and vegetables exceeds the standard.
