摘    要:在甾体生物催化过程中,向发酵液中添加亲水性有机溶剂,是增加底物溶解进而提高转化效率的有效手段,但有机溶剂对于菌体普遍具有毒性。为此采用N+注入诱变方法,对出发菌株雷斯青霉p-10-8进行改良,获得耐促溶剂菌株60 A-5-8,当投料量增大到4‰时转化率达82.61%。经过5次传代实验证明,该菌株具有较高的遗传稳定性。

Abstract:In the steroid biocatalytic process, adding hydrophilic organic solvent to the fermentation medium is an effective method to increase the substrate solubility and the conversion efficiency, but organic solvents are often toxic to mycelium. To solve this problem, the Penicillium raistrickii p-10-8 was mutated by N+ ion implantation, and an organic solvent tolerant strain named 60 A-5-8 was obtained, of which the conversion rate reached 82.61% when the amount of substrate increase 4%0. In addition, the mutant strain shows high genetic stability even after 5 generations of cultivation.
