超声波破碎法提取发酵性丝孢酵母胞内脂肪酶的条件优化

摘    要:利用超声波对发酵性丝孢酵母(Trichosporon fermentans)进行细胞破碎,研究了输出功率、辐射时间、工作总时间、菌体质量浓度等因素对胞内脂肪酶活性的影响。通过单因素试验和正交试验,获得较高活性脂肪酶的提取条件:超声波每次辐射时间为6s(间歇时间5s),菌体质量浓度为25g/L,输出功率为550W,工作总时间为5.5min,在此条件下提取的脂肪酶活力达到128.46U/g。

Abstract:The disruption and lipase release of Trichosporon fermentans by ultrasonication was examined.The effect of ultrasound output power, duration of irradiation each time, total irradiation time and the cell concentration on the activity of lipase were investigated through the single-factor and orthogonal experiment.The lipase activity reached 128.46 U/g under the optimum extraction condition: duration of irradiation each time 5s(with a 5sinterval), the cell concentration 25g/L, output power 450W, total irradiation time 5.5min.
