两种5-氨基-3-巯基-1,2,4-三唑衍生物的合成及结构表征

摘    要:5-氨基-3-苄硫基-1,2,4-三唑和5-氨基-3-正丁烷硫基-1,2,4-三唑都是三唑并嘧啶磺酰胺类除草剂的中间体,以5-氨基-3-巯基-1,2,4-三唑(AMZ)、苄氯,正溴丁烷为原料,通过对AMZ的烷基化合成上述两种化合物。通过正交实验得到较好的合成工艺条件:5-氨基-3-苄硫基-1,2,4-三唑的合成,温度为60~70℃,反应时间为10min,氯仿的量为25mL时产率可达70.4%;5-氨基-3-正丁烷硫基-1,2,4-三唑的合成,温度为70~80℃,反应时间为1h,AMZ与正溴丁烷的摩尔比为1∶1.1时产率可达78.4%。通过红外光谱、核磁共振氢谱等表征了其结构。

Abstract:As important intermediates of triazolopyrimidine sulfonamide herbicide,the synthesis of 5-Amino-3-benzylthio-1,2,4-triazole and 5-Amino-3-n-butyllthio-1,2,4-triazole are investigated in this paper.5-Amino-3-mercapto-1,2,4-triazole(AMZ),benzyl chloride and n-butyl bromide are adopted as the starting materials.The two compounds are obtained by the alkalization of AMZ.The orthogonal tests are also conducted to get optimal reaction conditions.For the synthesis of 5-Amino-3-benzylthio-1,2,4-triazole,the optimal temperature: 60-70 ℃,reaction time: 10 min,volume of chloroform: 25 mL,yield: 70.4%.For the synthesis of 5-Amino-3-n-butyllthio-1,2,4-triazole,the optimal temperature: 70-80 ℃,reaction time: 1 h,molar ratio of AMZ and n-bromobutane: 1∶1.1,yield: 78.4%.The structure of final product is also characterized by IR and HNMR.
