玫瑰香韵烟草突变体代谢轮廓分析

摘    要:  背景和目的  利用甲基磺酸乙酯(EMS)对中烟100(ZY100)进行化学诱变,获得一个具有特征香韵的香气突变体(玫瑰香),为明确玫瑰香烟草突变体与ZY100在代谢产物和代谢途径方面的差异。  方法  采用GC-MS对玫瑰香突变体和ZY100烟芽的挥发性次生代谢产物和内在代谢产物进行分析。结合主成分分析(PCA)和偏最小二乘法-判别分析法(PLS-DA)筛选差异代谢物,并对差异代谢通路进行富集。  结果  (1)共检出挥发性代谢产物27种,包括醇类、醛类、酮类和烯烃类,突变体中挥发性物质种类多于对照,主要成分为反式-β-罗勒烯。(2)共鉴定内在代谢产物60种,包括氨基酸类、糖和醇类、有机酸类和其他次生代谢产物,筛选到差异代谢物27种。(3)玫瑰香突变体中与氨基酸合成、糖酵解、三羧酸循环、有机酸代谢、生物碱和萜类合成等相关的19条代谢通路发生极显著变化。  结论  差异代谢物和代谢通路的改变赋予了玫瑰香突变体特殊的花香香韵。

Metabolite profiling of tobacco mutant with rosaceous flavor note

Abstract:  Background and objective  Chemical mutagenesis of Zhongyan 100 (ZY100) was carried out by using ethyl methane sulfonate (EMS) to obtain mutant with characteristic rosaceous flavor note. The purpose of this paper is to clarify the differences in the metabolites and metabolic pathways between the rosaceous flavor note tobacco mutant and ZY100.  Method  The volatile secondary metabolites and intrinsic metabolites of the rosaceous flavor note tobacco mutant and ZY100 tobacco bud were analyzed by GC-MS. Principal component analysis (PCA) and partial least squares-discriminant analysis (PLS-DA) were used to screen differential metabolites and enrich the differential metabolic pathways.  Results  (1) A total of 27 volatile metabolites were detected, including alcohols, aldehydes, ketones and olefins. The volatiles which are mainly represented by trans-β-ocimene were more in the mutants than in the control. (2) A total of 60 intrinsic metabolites were identified, including amino acids, sugars and alcohols, organic acids and other secondary metabolites, and 27 differential metabolites were screened. (3) The 19 metabolic pathways related to amino acid synthesis, glycolysis, tricarboxylic acid cycle, organic acid metabolism, alkaloids and terpenoid synthesis were significantly changed in the rose scent mutant.  Conclusion  The changes of differential metabolites and metabolic pathways lead to the characteristic rosaceous flavor note of tobacco mutant.
