摘    要:以液体深层发酵培养的姬松茸菌丝体为原料,利用酶法提取姬松茸胞内多糖。通过单因素试验、正交试验及方差分析确定了复合酶法提取胞内多糖的最佳条件:pH 4.5、温度45℃、时间1.5 h、木瓜蛋白酶1.0%、纤维素酶0.5%、果胶酶1.0%。在此条件下,胞内多糖的提取率为3.70%。胞内多糖经Sephadex G-200柱层析后得到两种多糖组分,经测定此两种多糖组分的相对分子质量分别为91 000和40 500。

Abstract:The intracellular polysaccharides of Agaricus blazei Murill by submerged culture were extracted by enzyme hydrolysis method.By single factor tests,orthogonal experiments,and variance analyses,the optimum hydrolytic condition is determined as follows: pH value 4.5,temperature(45℃,) time 1.5h,papain 1.0%,cellulase 0.5%,and pectinex 1.0%.Under this condition,the yield of intracellular polysaccharides is up to 3.70%.Two kinds of polysaccharides are obtained by Sephadex G-200 column chromatography with molecular weight 91000 and 40500,respectively.
