摘    要:在pH 3.5的Britton Robinson(B R)缓冲溶液中铬天青S与蛋白质能够发生相互作用形成生物超分子复合物 ,使铬天青S在 - 0 92V (vs.SCE)处的伏安还原峰峰电流下降而峰电位基本保持不变。优化了结合反应条件和电化学测定条件。在最佳条件下 ,峰电流的下降值同人血清白蛋白 (HSA)的浓度在 6.0~ 2 0 .0mg·L- 1 范围内呈线性关系 ,线性回归方程为Δip″(nA) =49 0 4 + 1 0 2 3c(mg·L- 1 ) ,γ =0 9962。将该方法应用于实际人血清样品的测定 ,结果与经典的考马斯亮蓝G 2 5 0光度法一致。

Abstract:In Britton-Robinson (B-R) buffer solution of pH 3.5, chromazurol S has a well defined second order derivative linear sweep voltammetric wave at -0.92 V (vs.SCE). Adding human serum albumin (HSA) to the chromazurol S solution resulted in the decrease of the peak current and without the change of peak potential, which indicated that a super-molecular complex was formed. The conditions of interaction and electrochemical determination were optimized carefully. Under the optimum conditions, the peak current of chromazurol S decreased proportionally to the HSA concentration from 6.0 to 20.0 mg·L -1 ,the linear regression equation is Δi p″(nA)= 49.04+10.23c(mg·L -1 ),γ = 0.996 2. The method was applied to determining the content of HSA in blood samples and the results were in good accordance with the traditional coomassie brilliant blue G-250 spectrophotometric assay.
