Abstract:Adding trace metal ions while the production of vancomycin fermented from Amycolatopsis orientalis V-0704 was optimized.Two level design for the trace metal ions showed that Co2+ and Mo6+ were the main influential factors for the production of vancomycin.The fractional factorial design,steepest ascent design and central composite design were employed orderly to optimize the chemical defined medium for the production of vancomycin by Minitab software.Finally the composition of the trace metal ions was obtained as follows: Co2+ concentration of 2.486×10-5g·mL-1,Mo6+ concentration of 2.486×10-5g·mL-1.The vancomycin yield adding the optimized trace metal ions was 4946 U·mL-1,which was increased by 17% compared with non-adding metal ions.
