摘    要:为了得到寇氏隐甲藻突变株的最佳发酵条件.先通过单因素试验考察接种量、培养时间、培养温度、装液量、培养基初始pH值等因素对二十二碳六烯酸产量的影响,在此基础上选择接种量、培养基初始pH、装液量为主要影响因素,通过Box-Behnken设计,得到最优发酵条件为接种量9.2%,初始pH值为6.9,装液量为65 mL/500 mL时,DHA产量最大为5.81 g/L,优化后DHA的产量比优化前提高了13.28%.

Optimization of fermentation conditions of Crypthecodinium cohnii for DHA production with response surface methodology

Abstract:To obtain the optimal fermentation condition of Crypthecodinium cohnii,single factor test was used to investigate the effect of inoculum,culture time,culture temperature,liquid volume,and medium initial pH on DHA yield.On the basis of single factor test,factors ofinoculum,medium initial pH,and liquid volume were selected as the main influence factors.Through the Box-Behnken design,the optimal fermentation conditions were acquired as follows:inoculum 9.2%,initial pH 6.9,liquid volume 65 ml/500 ml.Under such condition,the yield of DHA was 5.81 g/L,which was 13.28% higher than before optimization.
