响应面法优化酵母菌产胞外多糖培养基的研究

摘    要:对能够影响酵母菌胞外多糖产量的基础培养基成分(碳源、氮源、无机盐离子)进行单因素试验,在单因素试验基础上,利用响应面法对主要影响因素进行优化,得出最适培养基配方为蔗糖6.0%、胰蛋白胨0.2%、硫酸铵0.1%、硫酸镁0.035%和磷酸二氢钾0.1%.此培养基在pH值为6、装液量50 mL/250 mL锥形瓶、温度28℃、转速170 r/min的条件下,在第8天酵母菌YF01-g胞外多糖产量最大,为4.672 g/L.

Abstract:The medium composition (carbon sources,nitrogen sources,mineral) which can affect yeast extracellular polysaccharide production was optimized by single factor experiment.On the basis of single factor experiment,the main influence factors were optimized by response surface methodology,and the production of polysaccharide was determined by phenol sulfuric acid method.Results showed that the optimum medium formula was sucrose 6.0%,peptone 0.2% and ammonium sulfate 0.1%,magnesium sulfate 0.035%.Under the condition ofpH 5,liquid volume 25 ml/250 ml,temperature 28 ℃,rotate speed 170 r/min,the yeast extracellular polysaccharide production was the maximum of 4.672 g/L at the eighth days.
