栀子苷的精制纯化研究

摘    要:应用6种大孔吸附树脂进行静态实验,筛选得到最佳树脂;通过动态实验确定最佳树脂对栀子苷纯化的最佳工艺参数。结果显示,D1300树脂为最佳树脂,其最佳工艺条件为上样液栀子苷废液吸光度值A238nm为1.683 0,吸附流速1.5 mL/min,洗脱流速2 mL/min,梯度洗脱,2 BV水洗,4 BV体积分数为30%乙醇洗脱。收集体积分数30%乙醇洗脱液,浓缩干燥后得到纯度为82.23%的固态栀子苷产品。

Abstract:Application of 6 kinds of macroporous adsorption resin was studied by static experiments, and the optimum resin was screened. The optimum process parameters on purification of geniposide were determined by dynamic experiment with the screened resin. Result showed that the D1300 was the optimal resin, and the optimum conditions were geniposide waste liquid absorbance 1.683 0 under wavelength 590 nm, adsorption velocity 1.5 ml/min, elution velocity 2.0 ml/min. The gradient elute condition was 2 BV water elution and 4 BV 30% ethanol elution. 30% ethanol eluant was collected, concentrated and dried, and the geniposide product with the purity of 82.23% was obtained.
