摘    要:品系为"大雪青"的大丽花幼枝,经兰州重离子加速器提供的80MeV/u 12C6 辐照后,种植在甘肃省定西市临洮新兴花卉公司基地.辐照8Gy的幼枝有一株花色突变体,用随机扩增多态性DNA(Random amplified polymorphic DNA,RAPD)技术对突变体和野生型进行检测分析.琼脂糖凝胶电泳结果表明,所用的10条引物中,有7条引物共扩增出78条带,其中5条引物扩增出了11条多态性片段,从而在DNA水平上证实了两者之间存在着差异,为进一步探讨重离子诱变机理打下基础.

Abstract:Young shoots of Dahlia pinnata Cav (Daxueqing) cultivar were irradiated by 80MeV/u 12C6 ions. The as-irradiated shoots were planted in the field of Lintao Flower Corporation in Dingxi City, Gansu Province. Flower color mutant was induced by 8Gy irradiation. Mutant and wild type of Dahlia pinnata Cav were detected for their DNA level differences using RAPD (Random amplified polymorphic DNA) method. The spectra of electropho- resis in agarose gel indicated that among 10 primers used, the 78 bands are produced in 7 primers. Among them 11 polymorphic bands are produced in 5 primers, which substantiated the differences in the DNA level between them. Mutation mechanism by heavy ion beams will be further studied on basis of the results.
