摘    要:隐甲藻油中含有34.72%的DHA,通过乙酯化、尿素包合富集,酶催化甘油解转化得到更高含量的DHA -甘油酯.通过对隐甲藻油碱法催化乙酯化,其酯化率达到92.60%;对得到的脂肪酸乙酯再经尿素包合分离浓缩,所得DHA -乙酯的含量达63.30%.运用酶催化将DHA -乙酯转化成DHA -甘油酯,通过实验优化,所得最优条件是:甘油与DHA -乙酯摩尔比1∶4,NOV 435酶加量2%(占总底物的质量分数),50℃抽真空( 100 Pa),DHA -乙酯转化率达到80.85%.研究结果表明,通过物理、化学和酶催化相结合能得到高含量的DHA -甘油酯.

Abstract:The Crypthecodinium cohnii oil containing 34.72% docosahexaenoic acid(DHA) was used as material,and the high concentration DHA glyceride was obtained by ethyl esterification,urea adduction fractionation,and enzyme catalyzed glycerolysis.By alkali-catalyzed ethyl esterification,the esterification rate reached 92.60%.Urea adduction fractionation was adopted to concentrate and purify DHA ethyl ester,and 63.30% DHA ethyl ester was obtained.And then the glycerolysis reaction conditions catalyzed by enzyme was employed to convert the DHA ethyl ester to DHA glyceride,and the reaction conditions were optimized as follows:molar ratio of glycerol to DHA ethyl ester 1∶4,NOV 435 dosage 2%,temperature 50℃,vacuum 100 Pa.Under the optimal conditions,the DHA ethyl ester conversion rate reached 80.85%.
