摘    要:优化大黄游离总蒽醌的提取工艺,为其在农业、医药、食品等方面的开发提供理论参数。在单因素试验基础上,选取提取时间、溶剂配比和液料比3个因素进行Box-Benhnken响应面法试验设计,对其提取工艺参数进行优化。在提取温度60℃,提取次数2次,提取时间2.11 h,溶剂配比0.43,液料比23.23时,大黄游离总蒽醌的理论提取率最高,可达1.021%。根据实际试验情况,将其参数修正为提取时间2.1 h,溶剂配比0.4,液料比23.2,提取温度60℃,提取次数2次,经验证此条件下大黄游离总蒽醌含量为1.017%,与理论值较为接近。采用Box-Behnken响应面法建立的游离总蒽醌提取工艺模型提取率高,并能很好地预测试验结果。

Abstract:The extraction technique of total free anthraquinone from Rheum was optimized to provide theoretical parameters for its exploitation in agriculture,medicament,foodstuff and so on.On the basis of single-factor experiments,the effects of different time,solvent match and ratio of water to material on the extraction yield of total free anthraquinone from Rheum were investigated with a three-factor Box-Behnken design.The extraction parameters were optimized with Response Surface Methodology.The results indicated that the conditions of 60 ℃,two of the repetition times,refluxing for 2.11 h,23.23∶1 of the ratio of water to material and 0.43 of solvent match were optimum,and the yield of total free anthraquinone could be up to 1.021%.According to conditions of actual experiment,the optimum extraction parameters were modified at 60 ℃,two of the repetition times,refluxing for 2.1 h,23.2∶1 of the ratio of material to water and 0.4 of solvent match.The validated experiment showed the yield of total free anthraquinone was 1.017 % under those conditions which was in consistency with the theoretic value.Extraction model of total free anthraquinone from Rheum established by Box-Behnken method can precisely predict the experiment results.In the best conditions deduced from the mode,the extraction rate of total free anthraquinone is high.
