Abstract:Objective:To study the ameliorative effect of Lactucopicrin on FFA-induced steatosis in HepG2 cells and explore its possible mechanism.Methods:The effect of Lactucopicrin on the viability of HepG2 cells was observed.HepG2 cells were induced and cultured with FFA to construct the steatosis model(NAFLD model in vitro).After 72h intervention with Lactucopicrin,the number of lipid droplets,the content of triglyceride(TG)and total cholesterol(TC)as well as the activity of superoxide dismutase(SOD),the content of reduced glutathione(GSH),malondialdehyde(MDA)and the expression of lipid droplet coating protein(Perilipin3)were measured.Results:Compared with the model group,Lactucopicrin intervention significantly reduced(P<0.05)the number of lipid droplets,TG and TC contents,and increased considerably(P<0.05)SOD and GSH levels,and significantly decreased(P<0.05)MDA content and Perilipin3 expression levels in a dose-dependent manner.Conclusions:Lactucopicrin significantly reduced the number of lipid droplets,TG,TC,and MDA contents in FFA-HepG2 cells,increased SOD and GSH levels considerably,and effectively improved FFA-induced steatosis and oxidative stress in HepG2 cells.Therefore,its mechanism of action may be related to altering the process of lipid metabolism and alleviating the level of oxidative stress in the cells.
