流行性乙型脑炎病毒减毒株at222的生物学特性

摘    要:目的 分析乙型脑炎病毒减毒株at222生物学特性,并与亲代野生强毒株AT21进行比较。方法 at222株和 AT31株在伊蚊细胞C6/36上繁殖,培养上清用作病毒液,在Vero细胞上以病毒蚀斑法测定病毒滴度,以 小鼠脑内感染法测定其神经毒力。结果at222株在蚊细胞上生长缀慢,但仍可达到与其亲代强毒株相似的滴度, 即l08PFU/ml。在Vero细胞上形成蚀斑比其亲代野生强毒株AT31明显小,脑内感染小鼠不致死,而ADI株却表现 出了极强的神经毒力。结论 对乙型脑炎病毒减毒株出at222的生物学特性进行了全面分析,为研究和开发乙型脑炎 疫苗提供理论依据。

Abstract:Objective To analyze and compare the biological characteristics of attenuated Japanese encephalitis virus strain at222 and its parental strain AT31 .Methods AT31 and at222 strains were propagat-ed in mosquito C6/36 cells, and the virus tilers and neurovirulences of them in supernatant were detected by plaque assay on Vero cells and intracerebral test in mice respectively. Results at222 strain grew slowly in mosquito cells,however, the titer(108PFU/ml)of it was almost identical to that of its parental strain AT31 .The plaques of at222 strain on Vero cells were significantly smaller than those of strain AT31 .The intracerebral test in mice also showed low neurovirulence of at222 strain. Conclusion The biological characteristics of at222 stain are cmopletely analyzed in this paper.The study provides a basis in theory for developing JE vaccine.
