Combined quantitative detection technology for multiple tumor markers based on near-infrared fluorescent labeled protein chip

Abstract:In order to effectively reduce the mortality of malignant tumors and improve the universality, stability and sensitivity of tumor screening, a combined quantitative detection system for multiple tumor markers with near-infrared fluorescent-labeled protein chips has been established. The principle of near-infrared fluorescent-labeled protein chip immunoassay of the system and the biochip scanner used for protein chip scanning and analysis were studied. A double-antibody sandwich near-infrared fluorescence detection method was used. By coating a variety of tumor marker antibodies on the solid-phase carrier, the antigen corresponding to the tumor marker in the serum of the subject was captured. The reactant was first combined with biotin-labeled secondary antibody, and then combined with near-infrared fluorescent-labeled streptavidin. A specially developed biochip scanner was used for scanning analysis to obtain biological information on the content of tumor markers in the examinee''s serum. Finally, the detection system and Roche electrochemiluminescence equipment were used to detect 4 tumor markers in the serum of 10 samples. The experimental results show that the linear correlation coefficients of the detection results of the tumor markers CA125, CEA, CA19-9 and AFP by the two instruments are 0.995, 0.992, 0.991 and 0.990. The multi-tumor markers combined quantitative detection system of the near-infrared fluorescent-labeled protein chip can realize the simultaneous detection of multiple tumor markers for multiple persons, and the results are highly correlated with the results of Roche single-person single-index detection.
