伏马毒素B1新型荧光标记物制备及其在荧光偏振分析技术中的应用

摘    要:目的针对玉米中常见真菌毒素——伏马毒素B_1(FB1)开发新型荧光标记物,从而替代昂贵的6-(4,6-二氯三嗪基)氨基荧光素(6-DTAF)标记物,并在此基础上开发基于荧光偏振原理的伏马毒素快速检测技术。方法采用一锅煮的合成方法,以5-(4,6-二氯三嗪基)氨基荧光素(5-DTAF)为荧光基团制备伏马毒素荧光标记物,然后利用荧光偏振的原理建立基于5-DTAF的伏马毒素快速检测技术。结果基于5-DTAF的FB1荧光标记物与基于6-DTAF的FB1荧光标记物的荧光偏振分析方法具有相同的分析性能,IC_(50)值基本相等。结论采用5-DTAF成功制备了FB1荧光标记物,同时利用该荧光标记物开发了伏马毒素荧光偏振快速检测技术。该分析技术与基于6-DTAF荧光标记物的FB1荧光偏振速测技术相比,具有同样的分析性能,但分析成本降低了一半。

Abstract:Objective Preparation of new fluorescent tracer against common mycotoxins such as fumonisins B1, to replace expensive 6-(4,6-Dichlorotriazinyl) aminofluorescein (6-DTAF) marker, and development of technique for fumonisins B1 quick detection based on the principle of fluorescence polarization. Methods Using the one-pot method to synthesize 5-(4,6-Dichlorotriazinyl) aminofluorescein (5-DTAF) fluorescent marker, and then adopting fluorescence polarization technique based on 5-DTAF to detect fumonisins B1. Results Using 5-DTAF we synthesized new fluorescent marker of fumonisins B1, and developed fluorescence polarization technology for fumonisins B1 quick detection. Fluorescence polarization technology based on 5-DTAF and 6-DTAF fluorescent marker of fumonisins B1 has almost the same amplitude and IC50 value. However, the price of 5-DTAF is only half of 6-DTAF, so analysis cost is greatly reduced.
