一株嗜热毁丝霉菌株产纤维素酶条件优化

摘    要:以一株嗜热的毁丝霉H127-1为出发菌株,对其产纤维素酶的液体发酵条件进行了研究。结果表明,最佳碳源为稻草粉,最佳氮源为黄豆粉。当添加一定量的金属离子时,Fe^2+能显著促进H127-1产CMC酶以及β-葡萄糖苷酶,而Zn^2+和CU^2+则极大地抑制了其产酶;Ca^2+、Ba^2+和Mg^2+都能促进H127-1产CMC酶,但在一定程度上抑制了其β-葡萄糖苷酶的分泌。发酵液初始pH值、装液量、接种量、发酵时间对H127-1的产酶也有一定影响,最适宜条件为初始pH4、装液量30~50mL/250mL、接种量6%~8%、最佳发酵时间为2~4d。

Abstract:The cellulase-producing conditions of a thermophilic Myceliophthora sp.H127-1 strain were optimized as follows: the best carbon source was rice straw powder and the best nitrogen source was soybean meal,the addition of Fe2+ could remarkably improve CMCase production and β-glucosidase production but the addition of Zn2+ and Cu2+ would inhibit greatly CMCase production and β-glucosidase production,the addition of Ca2+,Ba2+ and Mg2+ in culture medium could improve CMCase production but would inhibit β-glucosidase production,the optimum initial pH value was 4,liquid medium volume was 30~50 mL/250 mL,the inoculating quantity was 6 %~8 %,and the optimum fermentation time was 2~4 d.
