副干酪乳杆菌产γ-氨基丁酸的发酵培养基优化

摘    要:以提高γ-氨基丁酸(GABA)产量为出发点,对具有益生特性的副干酪乳酸菌M5进行培养条件优化。采用单因素、PB与响应面试验对初始培养基配方进行优化。优化后最适培养基配比为:酵母浸粉10 g/L、无水乙酸钠2 g/L、蛋白胨5 g/L、葡萄糖10 g/L、L-谷氨酸钠1.4 g/L、维生素B6 2 mmol/L、氯化钙1.5 mmol/L、接种量4.1%。采用此培养基较初始培养基合成GABA的产量提高了9%。研究结果对提高副干酪乳杆菌产GABA能力提供了依据。

Abstract:To improve the ability of gamma-aminobutyric acid (GABA) by Lactobacillus paracasei, the L paracei M5 with potential probiotic characteristics was optimized in this study.Single Factor experiment, Plackett-Burman experiment and Response Surface experiment were used to optimize production for L paracei M5 on the basis of Initial medium.The optimal medium composition was determined as follows: the yeast extract 10 g/L, anhydrous sodium acetate 2 g/L, peptone 5 g/L, glucose 10 g/L, L-glutamate sodium 1.4 g/L, vitamin B6 2 mmol/L, calcium chloride 1.5 mmol/L, inoculation amount 4.1%.The yield of GABA synthesized from the optimized medium was 9% higher than that of the original medium.The results provide a basis for improving the GABA production ability of Lactobacillus paracasei.
