羟基磷灰石/丝素蛋白纳米复合颗粒的制备

摘    要:用氢氧化钙与磷酸湿法合成羟基磷灰石,加入丝素蛋白以诱导羟基磷灰石晶体的定向生长,以仿生的方法得到复合颗粒。通过扫描电子显微镜(SEM)、电子探针分析(EDS)、X-射线衍射(XRD)、红外吸收光谱(FT-IR)、粒径分析仪等检测手段,探讨了制备工艺条件对羟基磷灰石丝素复合粉体制备的影响及其合成机理。结果表明,制备的复合颗粒为纳米级粉体,长度在100-400nm,宽度在30-80nm之间。丝素蛋白可以诱导羟基磷灰石形成针状晶体,晶体的长轴方向沿着C轴方向,这是因为丝素蛋白与羟基磷灰石之间的相互作用造成的。并且随着丝素蛋白加入量的增加,长径比增加,随着温度的增加,结晶度增加,其组成和结构与入骨组织中纳米微晶非常相似。由于羟基磷灰石丝素蛋白复合纳米粒子与入骨中磷灰石微晶的相似性以及基体材料的可降解性,这些材料被赋了;优异的骨诱导性能和可降解性能,在骨修复或骨固定材料方面有着潜在的用途,可以为适合于临床应用的HA产品提供优质的粉体原料。

Abstract:Complex Hydroxyapatite(HAp)nanocrystals were prepared by coprecipitation method from Ca(OH)2 and H3PO4 using bio mimetic process,in the presence of silk fibroin(SF).Scanning electron microscopy(SEM),electron probe microanalysis(EDS),X-ray diffraction(XRD)and FT-IR were employed to discuss the effects of process conditions on the preparation and synthesized mechanism.The results indicate that the complex powers are nanometer,the length is 100~400 nm,and the width is 30~80 nm.Silk fibroin can induce preferential alignment of HAp crystallites along the longitudinal direction of C-axis,for the chemical interaction between HAp and SF.The complex powders have a poor crystallization and are similar to the natural bone apatite.Owing to the similitude of microcrystalline with human bone and biodegradation of radical materials,the Hydroxyapatite fibroin nanometer complex powders endow with excellent osteoindutive and bio degradation,which will be good candidate as a starting material for bone substitutes,and be a high-quality raw powder materials in clinic application.
