油茶皂素的提取及纯化新方法

摘    要:采用乙醇为浸提剂提取油茶籽饼粕中的油茶皂素,正交实验结果表明,油茶皂素提取的较优工艺条件为:乙醇浓度为50%,浸提温度60℃,料液比1:10,浸提时间2h,在此条件下油茶皂素的得率为16.66%,粗产品的油茶皂素的纯度为62.37%。用高速逆流色谱法对粗品进行分离纯化,初步的实验结果表明:乙酸乙酯:正丁醇:水(1:1:2,V/V/V)是适合于油茶皂素分离纯化的溶剂系统,能将油茶皂素含量提高到91.8%。

Abstract:Tea saponin was extracted from seeds of camellia oleifera with alcohol as extraction solvent.The results of orthogonal test showed that the optimum technological conditions were as follows:ethanol concentration 50%,soaking temperature 60℃,ratio of liquid to solid 1:10 and soaking time 2h.Under the conditions,the tea saponin yield was 16.66%,and its purity in crude product was 62.37%.Then the crude tea saponin was further purificated and separated by HSCCC.The preliminary results showed that the solvent system of ethyl acetate-n-butanol-water(1:1:2,V/V/V)was suitable for the purification of tea saponin and the content can be increased to 91.8%.
