摘    要:主要研究了水酶法提取松子蛋白的酶解工艺。选用Alcalase碱性蛋白酶作为水解酶,以总蛋白提取率为指标。通过单因素实验和响应面实验,得到影响实验的因素依次为加酶量>酶解温度>料液比>酶解pH>酶解时间。确定最佳的酶解参数为:加酶量1.9%,温度55℃,酶解时间2.2h,料液比1∶5,pH8.8,此时总蛋白提取率为87.92%。

Optimization of aqueous enzymatic extraction conditions of pine seed protein by response surface method

Abstract:Hydrolysis process of aqueous enzymatic extracted pine seed protein was studied. Alcalase was selected to extract protein from pine seed,and the extraction rate of total protein was used as index. Single factor experiment and response surface experiment were applied to analyze factors. The contribution rate of factors were as follows:enzyme amount>hydrolysis temperature>the ratio of solid to liquid>pH>hydrolysis time. The results showed that the optimum conditions were:enzyme amount 1. 9%,hydrolysis temperature 55℃,hydrolysis time 2. 2h,the ratio of solid to liquid 1∶ 5,pH 8. 8,the total extraction rate of pine seed protein could reach 87. 92%.
