低温碱性脂肪酶产生菌的筛选及产酶培养基的优化

摘    要:以橄榄油为唯一碳源,从渤海湾盐碱地被油污染的土样中分离筛选出1株产低温碱性脂肪酶菌株34-5,初步酶学性质研究表明,该菌所产脂肪酶的最适温度为25℃,最适pH为9.6,该菌的16S rDNA基因序列与伯克霍尔德氏菌(Burkholderia)的同源性为99%摇瓶实验表明,该菌株最适产酶培养基为(g/L);淀粉10,黄豆饼粉20,玉米浆20,K2HPO4 1,聚乙烯醇大豆油乳化液20.其最高酶活为6.87U/mL。

Abstract:A bacterium which produces cold-adapted alkaline lipase was isolated from soil on a selective medium that contained olive oil as the only source of carbon. The soil was collected from bay of Bohai. The optimal temperature and pH of the lipase were 25℃ and pH 9.6, respectively. The sequence of 16S rDNA of strain 34-5 showed 99% homology to Burkholderia. The optimal medium for lipase production were determined as follows (g/L) :starch 10, bean flour 20, corn syrup 20, K2HPO4 1, emulsioned bean oil 20. The alkaline lipase activity reached maximal level of 6.87 U/mL.
