摘    要:氯苯是重要的化工原料,长期使用对环境造成了严重的危害.为了研究生物方法修复自然环境的可行性,采集化工厂排污口的污泥,利用富集驯化等方式,分离到一株能够降解氯苯的菌株KD131.通过分析该菌株的16SrRNA基因序列,并结合BIOLOG微生物鉴定系统的测定结果,确定KD131为短小芽孢杆菌(Bacillus pumilus),同时构建了系统进化树.利用气相色谱方法,对分离菌株KD131降解氯苯的能力进行了初步分析,24h内氯苯分解率达60.8%.进一步研究发现,KD131对苯和邻二氯苯也具有一定的降解能力.

Abstract:Chlorobenzene is an important chemical material with many applications. In order to investigate the possibility of bioremediation of chlorobenzene-contaminated land,a chlorobenzene degrading bacterium KD131 was isolated from the effluent and sludge samples of a chemical plant. Minimal medium supplemented with chlorobenzene as sole carbon source was used during the enrichment and domestication process. 16S rRNA gene of KD131 was amplified and sequenced for identification. Combined with biochemical analyzing results with BIOLOG, strain KD 131 was classified as Bacillus pumilus. Gas chromatography was used to quantify the bio-degradation rate of chlorobenzene of strain KD131. Within 24 hours,60.8% of chlorobenzene was degraded in the minimal medium. Further experimental results also showed that KD 131 could degrade benzene and o-dichlorobenzene.
