摘    要:目的 依据国家标准样品定值要求,为提高标准样品纯度,降低杂质残留量,选择最佳栽培方式的灵芝子实体用于灵芝多糖肽(Ganoderma lucidum polysaccharide peptide, GL-PPSQ2)标准样品的研制。方法 基于高效液相色谱法(high performance liquid chromatography, HPLC),采用面积归一法对GL-PPSQ2主成分进行定量分析,卡尔费休库伦法(K-F)测定水分,热重分析法(thermogravimetricanalysis,TG)测定灰分,微波消解-电感耦合等离子体质谱法(inductively coupled plasma-mass spectrometry, ICP-MS)测定14种微量元素,顶空-气相色谱法(headspace-gas chromatography, HS-GC)测量溶剂残留;质量平衡法对菌草层架(A)、菌草覆土(B)、苡仁壳层架(C)、短段木覆土(D) 4组不同栽培方式所得GL-PPSQ2进行定值。结果 GL-PPSQ2定值依次...

Abstract:Objective According to the certification of national standards, Ganoderma lucidum(G. lucidum) fruiting body under the best cultivation conditions was selected for preparation of G. lucidum polysaccharide peptide,in order to improve the purity and reduce the residual amount of impurities. Methods The main component purity values of G. lucidum polysaccharide peptide was determined by high performance liquid chromatographic(HPLC) area normalization method, moisture was measured by Karl Fischer(K-F) method, ash content were determined by thermogravimetric analysis(TGA),14 trace elements were measured by microwave digestion-inductively coupled plasma mass spectrometry(ICP-MS)and solvent residue was determined by headspace-gas chromatographic(GC); the purity of G. lucidum polysaccharide peptides under four different cultivation conditions was analyzed by mass balance method. Results The purity of G. lucidum polysaccharide peptide(%): adlay husk layer frame cultivation(97.40)>Juncao layer frame cultivation (96.50)>Short-segment wood covering soil cultivation (96.08)> Juncao covering soil cultivation (94.40); The yields (%) of G. lucidum polysaccharide peptides were as follows: Juncao cover soil(0.29)>Short section wood cover soil(0.18)>Juncao layer frame(0.15)>adlay husk layer frame(0.071). Conclusion According to the experimental results, short-segment wood covered with soil and Juncao layer frame cultivation should be selected as the raw material for preparation of G. lucidum polysaccharide peptide standard samples.
