卡那霉素键合固定相的制备及手性拆分研究

摘    要:将氨基多糖类抗生素卡那霉素(Kanamycin)键合在环氧硅胶上,作为高效液相手性固定相(HPLC-CSP)并被应用于HPLC手性拆分.通过对卡那霉素CSP的制备、表征及在正相条件下对异博定、黄皮酰胺、萘乙醇等三种物质的手性拆分研究,探讨其手性拆分机理.研究结果表明,卡那霉素键合CSP对含有羟基、氨基、羰基等极性基团的手性物质在正相条件下有较好的分离能力.结论:氢键作用是手性分离机制的最主要的因素,流动相极性对溶质的保留和拆分有一定的影响,极性增加,保留减小且分离因子减小.此类CSP极性很强,在反相下会失去活性.

Abstract:A chiral stationary phase(CSP) was prepared by bonding the glucidamin antibiotic Kanamycin on epoxy silica(Kanamycin CSP) and was used in the chiral separation in high performance liquid chromatography(HPLC).The preparation,characterization and chiral separation of kanamycin-CSP in normal phase were discussed.There are chiral substances1-(6'-Methoxyl naphthyl)ethanol,Verapamil,clausenamide] that have been separated.The results show that kanamycin CSP can be used in the separation of chiral compounds which contain polarity group such as hydroxide,amido and carbonyl in normal phase HPLC.The conclusion is that the action of hydrogen bond is the uppermost mechanism of chiral separation.The polarity of mobile phase may affect the retention time and the resolution of chiral substance in canamycin-CSP,and the increased polarity will result in shortening the retention time and reducing the separation factor.This CSP has strong polarity with no reactivity in the reversed phase HPLC.
