摘    要:采用正交设计和双灭活标记方法对冬虫夏草与蛹虫草进行原生质体融合研究,获得融合子。结果表明:最适融合条件为:40% PEG 与0.08mol/L Ca2+ 的混合溶液(pH7.0)助融,35℃融合20min,融合率为2.74 × 10-5;并用形态学、颉颃实验及分子生物学方法初步探讨了双亲和融合子三者之间的亲缘关系。

Abstract:Fusion between protoplasts from Cordyceps sinensis and Cordyceps militaris subjected to heat and ultraviolet inactivations, respectively, was studied. In order to maximize fusion rate, five crucial fusion conditions were optimized using orthogonal array design. A maximum fusion rate of 2.74 × 10-5 was achieved through 20 min fusion at 35 ℃ under the promotion of 40% PEG and 0.08 mmol/L CaCl2 (pH 7). Besides, the genetic relationships among the parent and fusant strains were quested by morphologic observation, rivalry experiments and RAPD-PCR identification.
