生物发酵产物中辅酶Q_(10)的快速分离柱高效液相色谱法测定

摘    要:研究了用快速分离柱高效液相色谱法测定生物发酵产物中辅酶Q10的方法。发酵法得到的样品经匀浆后用四氢呋喃提取,提取液以ZORBAX Stab le Bound(4.6×50 mm,1.8μm)C18快速分离柱为固定相,m(甲醇)∶m(四氢呋喃)=4∶1为流动相分离,流速为2.0 mL/m in;在该色谱条件下,辅酶Q10在2.0 m in内可达到基线分离;用紫外二极管矩阵检测器检测。方法标准回收率为96%~102%,相对标准偏差为0.78%~1.22%。方法用于几种发酵样品中辅酶Q10的测定,结果满意。

Abstract:A rapid column high performance liquid chromatography method for the determination of coenzyme Q_(10) from fermentation was studied.Coenzyme Q_(10) was extracted from the sample with tetrahydrofuran by ultrasonic extraction and then separated on a ZORBAX Stable Bound(4.6×50mm,1.8μm) rapid chromatographic column with methanol-tetrahydrofuran(80∶20) as mobile phase at a flow-rate of 2.0ml/min,and monitored with the photodiode array detector at 275nm.The recovery was 96~ 102%,and the relative standard deviations was 0.78~ 1.22%.This method was applied to the determination of coenzyme Q_(10) from fermentation with good results.
