摘    要:以花生根白藜芦醇提取液为原料,对选取的5种大孔树脂进行静态吸附试验,确定DA-201树脂为最优吸附树脂.通过DA-201树脂吸附白藜芦醇的动力学试验、DA-201树脂等温吸附试验、上样量试验与动态洗脱试验以及考察上样流速、洗脱流速和洗脱溶剂浓度的三元二次通用旋转组合设计柱层析试验等研究发现:DA-201树脂等温吸附白藜芦醇过程符合Langmuir和Freundilch方程.在上样质量浓度为0.7 mg/mL,上样液pH 3,上样体积为20 mL,洗脱体积为15 mL的条件下,进行DA-201大孔吸附树脂柱层析纯化试验,建立了大孔吸附树脂柱层析纯化白藜芦醇的数学模型.经回归与方差分析,对方程进行局部寻优得出:在上样流速1.00 mL/min,洗脱流速1.60 mL/min,乙醇体积分数75%,其纯化后白藜芦醇的得率为(80.13±0.01)%,经HPLC检测其纯度可以达到39.61%.

Abstract:Taking resveratrol extract from peanut roots as experimental material and through static adsorption experiments,DA -201 resin was selected from five resins as the optimal adsorption resin for peanut resveratrol.Through DA-201 resin adsorption kinetics experiments ,a DA-201 resin isothermal adsorption test,a loading volume test,a dynamic desorption test and column chromatography experiments on three factors (loading velocity,eluting velocity and concentration of eluting solvent) ,it was concluded that the DA-201 resin adsorption process for resveratrol conformed to Freundilch and Langmuir equation.Under the conditions of sample concentration 0.7 mg/mL,pH=3,sam ple volume 20 mL and elution volume 15 mL,a test of purification of resveratrol by DA-201 macroporous resin column chromatography was carried out and a mathematical model was established.Through regression analysis and variance analysis,the optimum conditions for the purification were drawn.Results: The obtained optimum purification conditions for peanut resveratrol by DA-201 macroporous resin column chromatography are sample velocity 1.00 mL/min,elution flow 1.60 mL/min and ethanol concentration 75%.Under these conditions the yield rate of purified resveratrol is(80.13±0.01 )% ,and the product purity measured by HPLC can reach 39.61%.
